Suma, no need to pay for MSSE advice: your computer must be ON for the automatic scan to work. I suggest you do it all manually once a week.

1. open Microsoft Security Essentials and click the 'update' tab, then click the 'update' button. Wait until the update is complete

2. click the 'home' tab and select type of scan. 'Quick' is probably good enough. Click 'scan now'.
this should take about half an hour.

I do it that way just before leaving for an errant. If it's going to be a 4 hour errant, do the 'full' scan instead.
