I STILL have not had the chance to read some of Ariad's published work but I have been in contact with Michael Gilman regarding a number of issues:

There's been little in vivo testing of the SH2-targeted compounds. The work is still progressing through the specificity tuning; the primary goal is to address osteoporosis and, since the animal model for this is quite involved, the company wants to be confident in the in vitro data prior to moving forward. Animal testing is said to begin soon. Clinical trials are obviously much further away but candidates may be screened as early as next year.

The gene therapy project appears to be at a more advanced stage of development: there's a good deal of data from animals indicating the Ariad compounds 'work'. Clinical application appears to be closer for this approach as well: among a host of compounds which are apparently effective in animals, the company is attempting to select the most efficacious. The Fas-based "suicide" system is farthest along: it is currently being manufactured for formal testing with the goal of progressing to clinical trials early next year.

This is a synopsis of the information provided by Michael Gilman. BTW, the people at Ariad do read this site and derive a good deal of entertainment, as distinct from insight, from most of the posts.
