Most startup biotechs do nothing but burn cash (zero revenue) until they either come up
with something or disappear, thus they are even worse than amzn by your measurements.
That doesn't make them worthless.

You cannot compare a biotech startup to AMZN. The biotech initially has no product to sell. It is an all or nothing venture. Selling books is not a new science. No R&D with out fruit is possible. It simply is marketing a commodity in a highly competitive market.

Most startups are not almost bankrupt and losing money. Most are compares making a great deal of money but need additional funding to put infrastructure in place to keep up with their growth.

Let me name a few that may no longer be startups but were extremely profitable the day of the IPO.

CSCO, CSCC, ASND, COMS, WMT, BKS, BGP, S, JCP, STO, CS, etc.
