Welcome to the thread. If your mouse clicks are flaky it's easy to overlook the easiest solution. Have you unplugged and plugged your mouse back in? It can work itself loose. Your mouse driver could have gotten corrupted. Uninstall it and reboot and let windows reinstall it for you. If neither suggestion helps, we'll dig deeper. If you're running XP you need to turn System Restore off to prevent reinfection of your system. You can turn it back on after running another virus scan.
