For extra credit, one can try to figure out what will be needed to add the "fail-safe" feature to the above (not sure it would make sense if simple withdrawal of the dimerizing drug would effectively shutdown expression, if the system is not "leaky").

FWIW...From what I can recall, @ Keystone they had a poster using Argent exvivo (fuzzy memory, notes @ lab : )...I think they incorporated a bump/or hole (can't remember) in the binding domain, so when the drug is not present... transduction does not occur, and the hole/or bump prevents leaky transduction?
