Query genes for Iconix assay construction may be from diverse
sources including human, animal, plant, viral or insect genomes.
Using Iconix technologies, these genes are inserted into and
expressed in host cells in which they are not typically expressed in
nature. Iconix primarily utilizes yeast and mammalian cells as
hosts. Iconix also uses DNA microarrays, computational tools,
promoter traps and genetic manipulation to analyze the specific
and nonspecific genomic impact of query gene expression on the
host cell genome. This transcriptional profile, in addition to
potential query gene-induced physical changes in the host cell, is
used to empirically guide construction of an assay based on the
observed activity of the query gene in the host cell genome. This
observed activity is referred to as a phenotype. Iconix refers to the
resulting screen-ready cells as Activity Reporter Cells, or ARCs.
