NuCleave™ is a novel DNA analysis system based on cleavage of nucleotide analogs incorporated during DNA amplification. Typically, one of the four dNTPs normally used during amplification is replaced by an analog, resulting in complete substitution by the analog. The amplified product is then subjected to a proprietary chemical treatment that breaks each DNA strand at each occurrence of the analog, and the resulting fragment mix is analyzed by MALDI-TOF Mass Spectrometry. This procedure allows SNP scoring on both DNA strands and also provides information about the sequence surrounding the SNP. Applications of the method, which has been implemented in a highly automated format, will be described.
