Robert: Let's switch gears and talk about pre-processing. Are there ways that you
can pre-process and separate out collections of peptides or proteins to enhance the
signal?

Dr. Aebersold: Yes.You cannot take a proteome comprising several thousand proteins
and simply infuse the mixture into a mass spectrometer and hope that the mixture
would give you any useful information. You have to do pre-processing. Again, there are
basically two tracks. It’s either pre-processing on the protein level or pre-processing on
the peptide level, or some combination of both..............

A common implementation of peptide separation upstream of a mass spectrometer is
ion exchange chromatography followed by reverse-phase chromatography.


Jim Silverman and I were standing in front of the MLNM mass specs three years ago, getting a description of the HPLC interface.
