Cubist has bet the farm on transferase. I find it strange that a company that discusses genomics so much can have such a high portion of its effort directed at a single target, without a complete understanding of its heterogeneity. I like them, but they are too "early" for my taste. Hopefully they will hit on transferase with one of their two remaining partners.

I feel strongly that MCDE is taking far more promising and diversified approaches. They have limited programs to find variants of existing antibiotics and to inhibit the efflux mechanism (partnered with *the* company that has most to gain). They have a massive screening project to identify new targets of interest. I am mega-impressed with their efforts to build libraries, in particular the natural extracts.

Anyone could pick, out of the literature, a couple of hundred fragile metabolic targets. However, the determining factors for success involve heterogeneity of the target and the ability to synthesize an inhibitor that doesn't muck with humans. For this reason, it is extremely risky to just pick a random target and design a screen. MCDE and CBST are as different as companies can be. It is remarkable that, at first blush, they can sound so similar.

Rurka is intense and focused. I've not yet met Bostian, but I'm damn well looking forward to it; his company hums. Nobody can predict success in pharmaceuticals, but MCDE is a class act.
