They are all critical.

Phase one involves a few patients and looks at dosage and side effects.

Phase II involve a few more people and focuses on the above as well as efficacy.

Phase III looks at all of the above in a larger population to try and uncover side effects you would only uncover with a larger test population.

Most drugs (about 70-75%) that make it to the phase III stage are approved and marketed.
