Actually not all that surprising if folks would stop assuming/extrapolating peripheral immune system function to inflammation within the CNS. Macrophage and microglia are NOT the same and Mabs crossing the BBB is most likely to be, in most cases, a NON-neutral event. The level of stimulus required to trigger a MASSIVE immune-type/microglial response is unbelievably small. And if you examine the brain at a point in time too far on either side of an incredibly tight window, you are very likely to miss this inflammatory response: the microglia come AND THEN THEY GO AWAY, often leaving behind significant neuronal damage resulting from their release of cytokines, nitric oxide, and reactive oxygen species, etc.

So the drug companies claim their drug produces no significant inflammatory response in the brain and the eager investor corps sucks this up...rather than consulting those who know better (who would tell them how completely absurd this claim is based on the crude and superficial level of temporal and anatomical analysis permitted in human subjects).

A couple years ago a group headed by Catherine Whitacre in Cleveland progressed into human trials with an oral tolerizing approach to MS (using MBP I believe). The study had to abandoned in short order as a number of the patients manifested EXACERBATED symptoms.

As I've said in SI for years, I wouldn't touch a Neuro-BT for at least a decade, and I would NEVER base my investment in a Pharma or other BT on their neuro research.

For one simple reason: humans know squat about the brain (and we are perpetuating the misery of countless victims of degenerative and other progressive neurological diseases by foisting this lie about amelioration and/or cures).
