if I recall correctly - I am out of science now for almost 10 years -
p53 is a protein, which controls the cell cycle. When it is damaged, the cell cycle, i.e. cell division is running on and on. That means malign growth. Hence you have to take care that the correct protein is expressed again. There is a whole group of similar proteins coded for by the so called "tumor suppressor genes".
