Electrical recordings of ion channels provide detailed insight into channel function and effects of candidate drug compounds. A planar patch-clamp system - IonWorks APC - based on a silicon substrate enables fast and reliable voltage-clamp recordings from ion channels. Cells are positioned in a few seconds, providing seal resistances of up to several hundred G Ohm. Standard sample volumes are in the range of 10-50 microliter, strongly reducing the consumption of sample compounds. Using this approach, we have been able to measure ion channel activities on different cell lines in both cell-attached and whole cell configuration. The patch on a chip technique will open the door for a new dimension of pharmacological ion channel screening.<<

Histamine receptor activation elicits a biphasic calcium response, dependent on the concentration and duration of the agonist exposure to the cells. This response consists of an initial transient calcium flux from intracellular stores and a lower, prolonged calcium flux, from extra-cellular media. These phenomena were studied using Procel, a new fluorescence-based platform that performs automated 120-point dose-response curves and Schild plots, and which records data at user-defined, discrete incubation times following agonist exposure. Various cell lines endogenously expressing histamine receptors were used to study the effect of calcium removal and of various second messenger and calcium pump inhibitors on the biphasic response. Results demonstrate how Procel technology determines quantitative differences in Histamine receptor activation in terms of potency, efficacy, and signal pathway activation amongst cell lines.<<
