Peter- I have followed the small interfering RNA literature since the early "prokaryotic" days in which it was much better functioning compared to the several attempts in eukaryotic cells. There is still an issue with either technology in that po dosing has significant bioavailability problems. An analyst friend I know at Pacific Growth suggested to me about a year ago to look into ISIS' newer oligo technology once the "first generation" technology was finished (ie, all the trials had failed and the work in the clinic moved to the second or tertiary technologies). Though oral bioavailabitlity still sucks, there is much better experimental data with the latter approaches. The recent runup of ISIS since the last clinical failure could be explained by the now more recent oligo technologies moving forward.
It is my belief that siRNA technology is best for target validation, particularly when chronic treatment paradigms are needed. Of course there is room for IV formulations for acute care drugs IMO. The ISIS phosphatase target for type 2 diabetes (PTP1B) is very interesting in my view. I also would like to know more of the ELAN technology that apparently has increased the bioavailability of the oligos.

Gotta love this market for the biotechs, eh? What a month!
