profile difference between Estorra and Indiplon?

Well really there are two different Indiplons - the immediate release and the time release. The immediate release has a much shorter half-life than Estorra, and so is good for getting you to sleep, but not so good for keeping you asleep all night. The time release has an undisclosed profile at this stage.

What I meant by the possibility of a long tail is that usually it doesn't matter too much if a time release takes a bit longer than normal for certain people. Thus if you look at the drug level across a population after say seven hours, you might find some subset that still has an efficacious amount of drug on board- the tail of the distribution, so to speak. But for a sleep agent you want the drug level to be low enough when you wake up that there is no residual effect.

It's always very hard to compare across drug studies - by and large these drugs right now look fairly similar. In terms of next day effect, you are matching Estorra's longer half-life with the characteristics of the Indiplon time release. Estorra has a known defect - the metallic taste a fair percentage of people have when they wake up until they brush their teeth in the morning.

Long term my guess is that Indiplon will do better than Estorra, if only because of the PFE marketing muscle. But I believe both drugs will do very well.
