why else would some cronies be buying 48,000 Call Options before the announcement

That was a call spread (bought the 35's and sold the 37.5's). The person made $2.50 per contract less their cost, which was apparently about $0.50. So a very nice transaction, but one that would have paled into insignificance compared with just buying the 37.5's naked, which is what they would have done if they knew about an approvable with a long-term label.

Personally I owned the Jan 50 leaps - a very different play, but one that has ended up with about the same return as the call spread.
