From the limited reading i do in this area, the aspect that fascinates me the most (and likely confounds drug development) is the fact that activation of certain toll-like receptors induces a local response whereas others may not induce a strong local response but a more effective systemic one. So a class of drugs targeting this mechanism will probably require an extra level of research into modes of administration above and beyond the usual IV-versus-pill debate. As a rule, I don't like ODN drugs. But me not liking something shouldn't surprise anyone :-)
