I've followed POTP since they were controlled by SEPR and have owned POTP intermittently during this period (currently have a few shares) and posted about them a couple of times on the valuation thread.

I reduced my position sharply a while back when the early Rituxan combo trial results came out and the side effect profile looked much more severe than I was anticipating. If nothing else, the SE profile seemed to kill any prospects in Type II diabetes.

I haven't really been following them much since, and so I'm pleased to see this thread. Clearly still a very high risk, high reward play - be an easy 10-bagger if things work out well.
