Hsp90

The molecular chaperone Hsp90 is widely recognised as an exciting new target for the treatment of cancer and other diseases. Hsp90 is responsible for ensuring the conformational stability, shape and function of a selected range of key oncogenic proteins, including many kinases and transcription factors such as ErbB2, Raf-1, Akt/PKB, mutant p53 and many others. Hsp90 is overexpressed in cancer cells and is thought to be involved in various cellular processes, such as cell proliferation, differentiation and apoptosis. Inhibition of Hsp90 causes simultaneous blockade of multiple cancer-causing pathways by promoting the degradation of many oncogenic client proteins, thereby exhibiting broad-spectrum antitumour activity. Although the reason for therapeutic selectivity in cancer cells versus normal cells is still unclear, new research results indicate that Hsp90 in cancer cells exists in a heightened, activated state that is highly susceptible to inhibition.
