David, Here's another post from the diabetes discussion group and it begins to show why ERGO and AMLN (for type II) may have a tough time of it, even if approved. Pioglitizone is one of the second generation TZDs in advanced clinical trials. Its made by Tekeda, JNJ's new partner in the far east. Another TZD, BRL49653, is made by SBH, and its also in advanced trials. I think that BRL49653 is on a par with Pioglitizone. Rezulin produced a 3% drop in HbA1c and this poster thinks that Pioglitizone is "10 times better".
LGND used BRL49653 in its Nature study on mouse models of diabetes as well as the Harvard study for treating liposarcomas. Targretin and ALRT268 were are a par with BRL49653 for diabetes, and the rexinoids synergized with BRL49653 for the treatment of diabetes as well as liposarcomas.

Hi Ken- most Doc's start at the lowest doseage (in this case 200 mg) to see how YOUR system responds. I started out on 200mg & it was not effective-- I asked if anyone was experiencing any degree of success with that dose & there was not anyone on the list on a "Rezulin only" treatment who is.
We upped the dose to 400 & it's somewhat better, considering my exercise routine has bit the dust since nasty storms hit the Pacific Northwest = <
I did a 6 month drug study on another Troglitazone drug (Pioglitazone) that, as my Doc explained, is 10 times stronger than Rezulin. They were doing 15 & 30mg doses. I had excellent results with that particular drug, which yielded an A1c of 5.7, in the final test. This drug is in final FDA testing phases, and may be out on the market in '98 Already in use in Japan for a few years now.
