Dont think they really appreciated that back in 1989, you need good confocal microscopy methods to sort that out.

The whole cell BCECF techniques were certainly a step up from microelectrodes and weak acid/base distribution methods they replaces, but they could only report the average signal for the whole cell and investigators had to make critical assumptions that the fluorescent dye was exclusively in the cytosolic compartment under all experimental conditions. Intracellular calcium robes (Quin-2, Fura-2, Indol-2) have the same whole cell resolution problems. You should probably stop relying on references from old patent applications.

You are right, pH 7.0 is neutral...just like most of biochemistry 101 tells us the inside of actively growing cells should be...not 6.4 in the cytosol, at least....not for long
