Daniel thanks for your insight but I'm not sure if I can agree with all your comments.

<<What really struck
me is that Dr Crook, the ISIS CEO, is medically oriented (that sounds funny for a
biotech company, but when you think about it, most biotech are run either by PhD
with no medical background or by MBA, also without medical background).>>

Don't sell PhDs short. PhDs are the men and women that come up with things such as antisense etc... that MDs eventually use in the clinic. I think both are needed in a biotech company, as are people with a strong business sense (i.e. MBA), since after all we are investing in businesses.

<<Then how are the target cells
specifically...targetted? (All cell types absorb to some degree DNA floating around
in liposomes). In vitro, no problems, it is sufficient to add oligos to the cell medium.>>

I believe you have your information backward. In cell culture oligos are not taken up by cells and thus require liposomes. However, in vivo oilgos are taken up efficiently and thus do not necessarily require liposome carriers.

<<In vivo, the antisense concept remained to be supported.>>

Please read the numerous reviews by various scientists including ISIS scientists that clearly show high efficacy in decreasing tumor formation in experimental animals. ISIS has demonstrated human tumor inhibition by antisense targeted at the Ras gene in human tumor xenografts in nude mice.

Hope this helps. Good luck.
