Agouron is taking the stance that an MMPI (matrix metalloprotease inhibitor) does function, at least in part, as an anti-angiogenesis compound (http://biz.yahoo.com/prnews/980505/ca_agouron_1.html).

As to "created by rational drug design", one could argue that the X-ray crystal structures didn't help them get to AG3340 . . . which is immaterial to the patients or the investors but interesting to us chemists.
