Say you think Microsoft is due for a huge tank in the near term, and it's trading at about $100 right now. You would call your broker to place an order to sell short MSFT 1000 shares. What this means is that you borrow 1000 shares of MSFT from your broker's account and you sell it on the market. Say then that in a week's time, MSFT tanks to $80/share. You feel pretty confident that it won't go any lower, so you call your broker to "cover" your position. What that means is that you buy 1000 shares back at $80/share and return the stock to your broker's account. You have just made the spread ($100-$80) less commissions, fees, and borrowing costs. This means in this example that you grossed $20,000.00 Pretty good trade.
