Hi Rick,

(a few more online thoughts : )

From what I understand, most Ad vectors are made of the same proteins as the wt. The main difference is inside the viral capsid part of the viral DNA sequence is modified, some wt DNA sequence is deleted and replaced with your GOI (E1 and E3 or E4 deleted Ad vectors)(E1 starts the transcription cascade...E3 helps the virus evade the immune response...E4 I'm not sure)(E1 is supplied in trans by the 293 cells). There is also a helper dependent form of Ad vector that has a reduced immune response (Kohnosuke Mitani played a role in developing this system...he has a position @ UCLA)...where most of the Ad wt genome is deleted (replaced with your GOI) and the viral proteins are supplied in trans via a helper virus.

Personally, I'll pass on the tolerization ; )

Best of Luck,

Where'd He Go?
