Guess you'll need to re-read The Billion Dollar Molecule for the much longer and gossipier version of Casaubon's post.

Made it more frustrating that some of these proteins (cyclophilin at least; I am too lazy to check on FKBP12 et al.) have proline isomerase activity, which led people to believe initially that these were classic enzyme drug targets. Calcineurin and the rest of the story explained why nothing simple worked.

Thanks to Casaubon from this end too.
