Whole Genome Association Studies in Humans

Most human traits result from the complex interaction of genetic and environmental factors. We have designed a new genetic mapping tool that will accommodate genome wide association studies with unrelated individuals and thereby overcome problems inherent in traditional mapping strategies. The tool consists of a set of 60,000 ancestral biallelic markers (single nucleotide polymorphisms or SNPs) homogeneously distributed over the human genome. Mapping is carried out by exploiting linkage disequilibrium between the SNP marker alleles and trait-influencing alleles. We have performed "proof of principle" studies for this mapping construct by describing associations between SNPs on our map and known diseases and disease genes. We have also developed novel analytic methods and theoretical constructs that will help guide the use of our mapping efforts. Finally, we have been able to develop, for the first time, an investigative strategy to isolate genes that influence drug responsiveness from clinical trials data.
