My first target would be to check how many programs you have loading on start-up and decide if they are necessary for your computer. To do this click on 'start','run',type'msconfig',hit 'ok', and click on the 'start-up' tab,
This will show you a list of programs with check boxes next to them. Programs like real player, AOL instant messenger (ie extras) you can probably disable safely. Then check apply and if you want restart. Most programs also have configuration screens that have 'Load on start-up' enable by default. In most cases all this option does is make the program start faster however if its a program you never use then its a waste of memory.

For the programs you are unsure you can post them here or PM them to mark.
As to the slow down question I'm afraid thats outside my little piece of knowledge ..
