To manually back up the registry go to start/run/type in msinfo32 and click ok. From this window choose tools/registry checker and (unless you have disabled it) you will be prompted with a window telling you that the registry has already been backed up once today, would you like to back it up again? Choose yes and then see if you can solve your problem. If things don't go well you can restore the backup registry by choosing the restart in ms-dos mode option on shutdown. At the c-prompt type scanreg/restore and hit your enter key. You will be given a list of good configurations to restore from. Choose the latest one and hit enter. Word of warning....some programs you may have installed can prevent this restoration and require a different way of doing this. Playing with the registry can make your computer unusable.
