Small molecular size is less likely to have problems getting through the gut intact and less likely to excite an immune response. But small molecules give you less to work with to allow fitting to a receptor and unlike antibodies there isn't a whole bunch of naturally occuring machinery sitting around to aid the development of the receptor.
