As the largest class of cell-surface receptors, and the site of action for a large proportion of currently marketed drugs, G-protein-coupled receptors (GPCRs) are of substantial interest to researchers in both academia and industry. But for their potential as therapeutic targets to be fully realized, several questions about the biology of GPCRs need to be addressed. Here, we present the responses of 20 of the world’s leading authorities on GPCRs to 20 questions posed about the current state and future direction of the field.
