I think velocity has a lot to do with it. Those old slow heavy civil war bullets would sooner take your arm or leg off than pierce a hole through it. I would think the key to stopping power would be keeping the bullet big relative to the size of the charge. Make 'um feel like they have been kicked by an elephant. Otherwise they might not even notice that they have been hit until they fall over dead a few minutes later. But like you said you won't be waiting around between shots to see how they react to your first one...
