Yep, that's one significant part of it: "I believe any color of the number -- this size -- will be problematic within the existing shade of grey." The background grey is too "middle" so it won't offer a lot of contrast for many colour options. The nav strip should be rethought at that point and reduce some of the visual clutter from the "beveled" edges.

Another important visual cue: since this is where all mail is handled, not just "inbox", the link should be defaulted to read "mail" normally, and change to "inbox X" only when there's new mail in it. Should be fairly easy to do since there's already a text string being changed in the cell anyhow.
