To make it short, they said they consider Israel and America one and the same and they do not believe Israel would do anything without US permission. In other words, an Israeli attack will be considered as an attack by US.

So should we consider their threats of destroying Israel with nuclear weapons to be a threat against the US?

If they are going to threaten Israel with destruction Israel has the right to take measures to avert that destruction. Israel would be reluctant to do anything drastic without the acceptance of the US but if it felt threatened in a major way it would. It would not exactly be a wise idea for Iran to then turn and attack the US.
