PCYC had the same happen - there a fairly sophisticated matching algorithm against historical controls in their Phase II showed a very clear benefit that wasn't repeated in a double blind randomised Phase III (although they are trying again in a subset and so the story isn't over yet). Obviously at some point one has to believe - but these examples show that point is further than one might at first think. Peter
