"The role of metabolism in inducing apoptosis was studied further by the investigation of the stable metabolites of clozapine and an analog of clozapine that is refractory to metabolic bioactivation. In accordance with our previous study of clozapine-induced cytotoxicity (Williams et al., 1997), we found that demethylclozapine was less potent than clozapine in inducing apoptosis via the bioactivation-dependent pathway, probably reflecting a lower cellular uptake, because turnover to its nitrenium metabolite was similar to that of clozapine."
