Dauntless, e your post. More Phentol pharmacology.
For any bodu tha thas taken pharmacology it is well known and established that ALL drugs manifest a curvilinear blood curve to a greater or lesser degree.In other words as sthe drug is being absorbed from the stomach, a 1/2 hour to 45 min process, the blood levels slowly rise to a PEAK and then gradually fall as the drug is being metabolized. So for every drug there is a so called " peak level" and a " trough level". The curve is more, or less, curved depending on each drugs rate of absorption and metabolism. At the " peak level" is where you get hit with the greatest amont of drug. The peak and trough levels can also be determined approximately clinically by the effect of a drug on you: like taking Tylenol: it will work well for a couple of hours ( at it's peak level) and then it wears off to it's tough.
For phentolamine studies ( see Goodman and Gillman Textbook of Pharmacol ) long time ago have shown that the onset of peak and trough for phentolam are very rapid. That is why it is not usefull.
Theoretically you could meke a sustained release formulation
such as Contac- 12 hour cold pill, gg. The problem though with phentol is that would also prolong it's systemic vasodilatory effects and knock your blood pressure down and subject you to 12 hours of arrhythmias.
I am very soory that you find this subject boring and I fully sympathize with you.For me learning the 1000 + pages of Goodman and Gilman was not one of my happiest experiences either. I promise however to make every effort to become less boring in th future and try and present the matter at a level where you can understand it.It would help a lot if you got your self a copy of Good & Gill or the Amer Hosp Pharmacop and started reading up on it.
