Altering secretase activity is correlated with changed patterns of Aá deposition. Mutant presenilin activity is also crucial for APP cleavage to the 1-40 peptide. These are circumstantial factors, mostly pertaining to the small subset of FAD cases (e.g., non-sporadic) where the risk factors are better understood yet the underlying causal roots or constellation of events are unknown. Identifying the underlying mechanisms of AD leading to the organic loss is a monumental task; the Sibia/BMY secretase program examines one candidate participant that may or may not address the underlying organic loss.

A paper suggesting Tacrine has anti-Aá activty independent of its anti-AchE role.
