What first separated Humans from Chimps.

Research by Harvard Professor Daniel E. Lieberman suggests it was the ability to throw rocks.

To be more accurate, the ancient creature clade australopithecine was the ancestor to both humans and chimps, but the chimpanzee has not changed much from that appearance and mode of living while humans have changed quite a bit. The change happened primarily when some of the common tribe realized they could throw rocks at predators and they would leave their kill thereby leaving a bunch of meat behind. Some 3 -5 million years ago rocks of a size and shape that are useful for throwing start to be gathered and concentrated.

Some of these apes were better at throwing rocks than others. They were favored for their ability to gather more meat than the tree hugging cousins. Still molecular DNA evidence suggests that from 5 to 3 million years ago the rock throwers retreated up into the trees along with their arboreal cousins to sleep and do a bit of mating. Still, there were becoming specialists. Finally, about 3 million years ago some apes could throw well enough that they didn't have to retreat to the trees, they just kept a stash of rocks nearby, and they became much better at throwing at the expense of the ability to efficiently move about trees. Throwing required changes in the shoulder and arm that were not particularly suited to climbing trees.

These throwing specialists were now confined to living mostly at ground level, for which they would have to stand up if they wanted to look around. The changes continued. Eventually they threw other things.
