Primarily, it's based on speculation on available data such as siesmic. This data can't precisely give you a definitive amount. It's better to error on the lowside than the highside, but that is a bit misleading. It's presuming that there is a strucuture that physically contains the proper pororisty and permeability to support a resivoir. I've seen plenty of area's that predict the "potential" of containing X amounts of oil.

It all part of the long process of determining where to sink the money, er, drill the well.
