David; Headaches?, I will take a headache over earlier death any day. In any event subtle changes in the molecule might eliminate one side effect and keep the main effects intact.
I think people are missing the major potential drug in CT-2584. I read from New Scientist ( newscientist.com ) in the 29NOV, 1997 issue that this drug bypasses the usual multidrug resistant mechanism(MDM) where the survivor cells learn to activate cellular pumps that pump out toxic materials at 100 times the rate of normal cells. In secreting cells these pump genes are already working to put out what the cells make, and so after 1-2 rounds of chemotherapy the few cells that hyper-express those genes proliferate and soon make up the main mass of the tumor, which then completely resists chemotherapy.
CT-2585 stimulates and enzyme (phosphotidyl-choline-phospholipase-D)
This enzyme is found in tumors at 15 times the concentration of normal cells. When excessiveley stimulated it causes overproduction of phosphatidic acids. The acids cause the cellular mitochondria to burst(probably by severe imbalance of transport processes).
Normal cells are slightly stimulated, and the rapidly dividing cells of the intestine, bone marrow etc are also unaffected.

The beauty of this is you can repeat it again and again as there seems to be no selection based survival mechanism. Some patients have had 6 treatments and have had their tumors eliminated(five year rates?? too soon yet)

If this works and it does seem to work, it will be enormously successful.

I certainly hope it lives up to its promise.

Take your point, & thanks for the tip on lisofylline, but do you have any info on CT-2584, which is at an earlier stage of development, but sounds to have v interesting potential for anti tumor activity ?
