Inhibition of A[beta] Peptide Formation In Vitro and In Vivo

We have studied numerous compounds which alter APP catabolism in a manner resulting in reduced A[beta] peptide formation
in vitro. Some of these compounds appear to modulate [beta]-secretase activity, while others work by a mechanism consistent
with modulation of [gamma]-secretase. Recent studies involving distinct lines of transgenic mice have shown that some of these
molecules are highly effective at inhibiting the proteolytic generation of the A[beta] peptide from APP in vivo. These
experiments will be discussed in detail.
