Receiving of the Holy Spirit is vital to any persons walk with God. It's He who convinces us that we're sinners and points us to calvary to make things right. It's also He who quickens our understanding relating to the things of God.

As to the counting of the 50 days from the day of His sacrifice, I'm not familiar with that, but I have always enjoyed the significance of numbers because it helps to bridge the believers understanding between the Old and the New Testaments.

Moses spend forty years in the wilderness after escaping Egypt before God called him into the ministry and spent forty days in the mount receiving the law.
It was also forty days that the spies searched the promise land, but because of unbelief the children of Israel were caused to wander in the wilderness for forty days and forty nights. Now Jesus on the other hand was tempted in the wilderness for forty days and forty nights but thank God, He came out more than victorious. After His resurrection it was said in Acts that He was seen for forty days.

In both of these examples there are at least 4 things in common. A wilderness experience, a set period of time, temptations, and faithfulness. I believe there is much that can be gleaned from this.
