A significant portion of the mutual fund money pouring into the market goes into index funds. The managers of these index funds will now have to load up on ASND.

Yes. All of those S&P 500 index funds (and there are quite a few now), are obligated to buy them.

The question is, will profit taking compensate for the "artificial" rise? Likely yes. But how soon, and can we little guys profit from this action? Maybe sell in the morning, buy back in the afternoon.

Because some mutual funds hold all the stocks in the S&P 500, and they are therefore required to buy it to balance their index. It is a familiar theme, and there are actually people who do nothing more than try to predict which stocks are next to be added, etc...

If you are a trader and it goes up 20%+ sell it. You can buy it cheaper later.
