I thought the S&P 500 was weighted based on each company's relative capitalization. If so, ASND wouldn't necessarily comprise 1/500 of the index, would it?

Stilts, I think its unweighted. Market caps change constantly, and would require way too much management to be cost effective. These funds typically sport the lowest management fees of all funds, and are typically termed "unmanaged".
