Your questions seem very sensible to me. We would think at least 32-pairs of subjects/controls would be neither unreasonable nor cost prohibitive. I am of the opinion any experimental design should be a matter of public record; there in all procedures and variables (including time) should be tightly defined.

If no one has a clue when and if data is due, who's running the show?
