IL-1b and TNFa are produced locally within tuberculous granulomas (28,29), and are sufficient to induce formation of granulomas when coupled to inert particles (30). Neutralizing TNFa antibody administered to BCG-resistant mice inhibits the formation of bactericidal granulomas following BCG challenge, and results in progressive, lethal BCG infection (31).
