II Corinthians 12

7 And lest I should be exalted above measure through the abundance of the revelations, there was given to me a thorn in the flesh, the messenger of Satan to buffet me, lest I should be exalted above measure.
8 For this thing I besought the Lord thrice, that it might depart from me.
9 And he said unto me, My grace is sufficient for thee: for my strength is made perfect in weakness. Most gladly therefore will I rather glory in my infirmities, that the power of Christ may rest upon me.

A vast wealth of wisdom is provided for study in these few verses.
Paul speaks of pride, the enemy, appealing unto the Lord, God's grace and where His strength is evident.

Laying aside scholarly opinions of whether Paul was afflicted physically or oppressed spiritually by a demonic influence, look at what transpired.

Paul was praying for relief and did not give up of his own accord. After three petitions by prayer he was in fact instructed by God that His grace would suffice. It was not God's will to prevent what was happening to Paul and we are even given the reason as having been to prevent pride.

When praying for someone or for ourselves we may not always know God's will. A lack of an immediate answer does not mean that we are praying against His will but we must remember that our timetable is not necessarily God's.

Paul had the advantage of receiving a "no" but even in that there was a blessing as we can see in Paul's words.

There is a song with a lyric something to the effect of "Some of God's greatest gifts are unanswered prayers."
