Further reading about the difficulty in cancer trials and the trials that resulted in the FDA approval of Genentech's Herceptin:

All in all, a pretty good and quick read. Most people reading this thread realize that Herceptin is a humanized mAb that is successful in treating her-2/neu overexpressing breast cancers.
