Some correct me if I'm wrong. This is my understanding: First of all, NASDAQ trades are via market makers, while AMEX and NYSE use specialists. Secondly, by watching the realtime change in ask size and bid size one can determine whether a block was a sale or a buy. If there is transaction of 10k shares and the ask size falls by 10k, the 10k was a buy, 10k fewer shares available at the ask.
